Tissue is commonly diagnosed by an imaging approach called multiphoton microscopy. In this technique, the tissue is illuminated by a pulsed laser and its fluorescence and scattered light are detected and analyzed. Since fluorescence is generally the best way to image the inside of biological tissues, multiphoton microscopy is the most used technique for clinical diagnosis. However, standard imaging systems are limited to a field of view of about 1 mm2, and consequently they cannot image deep in tissue. In addition, multiphoton microscopy does not have an adequate spectral resolution to perform a complete imaging of the tissue. Here we describe a new multiphoton microscopy imaging technique, which allows for optical sectioning, spectral selectivity, and is capable of imaging deep in biological tissues. The principle of the technique is based on the use of vibrational transitions of molecular nitrogen-vacancy centers in diamond, which are modified to be sensitive to a particular biological marker (fluorophores) and to the laser excitation used to image the tissue. Using this technique, we were able to selectively image apoptotic regions in tumor tissues, or measure multiplexed spectral signals from the same spatial location in the tissue.
